چکیده

Previous work has demonstrated that the HbS gene has appeared and expanded three times in Africa in three separate geographic locations and that these three distinct mutational events can be identified by linked DNA polymorphic sites (haplotypes) surrounding the abnormal gene. We have reported that the Senegalese and Beninian haplotypes differ in G,., , expression. mean percentage of HbF. and percentage of dense cells. We now report on the third haplotype. the Bantu. and find that it has intermediate features. namely. the high mean percentage of HbF and low percentage of dense cells associated with the Senegalese haplotype. but with a low percentage of G expression similar to the Beninian haplotype. The distribution of

Level of Hemoglobin F and Gg Gene Expression in Sickle Cell Disease and Their Association with Haplotype and XmnI Polymorphic Site in South of Iran

Background: Molecular genetic factors regulating hemoglobin F (Hb F) expression are important modifiers of the severity of sickle cell anemia (SS). Methods: The prevalence of XmnI polymorphic site, the Gg:Ag ratio and the Hb F level were determined using PCR-RFLP procedure, HPLC and alkaline denaturation method, respectively, in various haplotypes of 52 patients with SS, 18 patients with sickle...

Beta-Globin Gene Cluster Haplotypes in Iranian Sickle Cell Patients: Relation to Some Hematologic

Background: Sickle cell anemia is relatively common in Khuzestan province located in Southwest Iran. The characteristics of sickle cell disease in Iran are apparently different from other regions some of these characteristics might be related to β-chain haplotypes. The purpose of this study was to determine the frequency of β-chain haplotypes in 50 patients with homozygous sickle cell anemia in...

A gene conversion located 5' to the A gamma gene in linkage disequilibrium with the Bantu haplotype in sickle cell anemia.

Cloning and sequencing of the gamma-globin gene of a sickle cell anemia patient homozygous for the Bantu haplotype has revealed a gene conversion that involves the replacement of an A gamma sequence by a G gamma sequence in the promoter area of the A gamma gene. This event is similar to another gene conversion believed to be responsible for the very high homology between gamma-globin genes, sug...

Results of Transcranial Doppler in Children with Sickle Cell Disease: Correlation between the Time-Averaged Mean of Maximum Velocity and Some Hematological Characteristics

Abstract Background: Transcranial Doppler ultrasonography results have not been previously studied in among Iranian sickle cell anemia patients. The present study was performed to evaluate the pattern of intracranial flow velocities among Iranian children with sickle cell anemia and the hematological parameters that can affect the time-averaged mean of maximum velocity in major intracranial...

Elucidation of βs/ Globin Gene clusters Haplotypes Related to Sickle Cell Anemia in Khuzestan Province, Southwest of Iran

Background & objectives: The researcher clarified that β/Globin gene cluster haplotypes in patients with sickle cell anemia provide useful population data as predictors of the disease severity, gene flow, and the origins of sickle cell mutation in this region. Materials and methods: A total of 150 subjects was investigated in two different groups for five polymorphism restriction site...
